Clinical development in immuno-oncology has unique challenges that make it difficult to position drugs effectively. Traditional pharmacokinetics (PK) and pharmacodynamics (PD) models often fall short in predicting the efficacy and safety of immunotherapies. The systemic and intratumoral immune effects do not consistently align with standard dose-toxicity-efficacy relationships that are well-understood in cytotoxic therapies.
